Effects of pear (Pyrus pyrifolia) methanol extracts on the production of nitric oxide and tumor necrosis factor-¥á in mouse peritoneal macrophages

Abstract
This study was conducted to investigate the effects of pear methanol extracts (P-M) on mouse peritoneal macrophages. Peritoneal macrophages were isolated from mice and cultured with or without LPS in the presence of P-M(0.16,0.31,0.63,1.25,2.50mg/m1) for 48hrs (TNF-¥á) or 96hrs (NO). NO levels in culture supernatant were determined by measuring the amount of nitrite, the stable oxidation product of NO, and TNF-¥á levels in culture supernatant were assayed by a sandwich ELISA. P-M significantly stimulated NO and TNF-¥á production from peritoneal macrophages at all doses. However, LPS-inducted NO production was inhibited by P-M in a dose dependent manner and LPS-induced TNF-¥á production was not altered by P-M.
